South Lanarkshire Council has said essential resurfacing works are to begin in Carluke.
Starting next week, works will commence along Mauldslie Road.
As a result, the road will be closed between Gasworks Road and Lawhill Road from Monday 29 July until Friday 2 August.
In order for the work to be carried out along the stretch of road, no vehicle access will be permitted during this period, except for those vehicles engaged in the works.
Councillor Chris Thompson, Chair of the Council's Enterprise Services Committee, said: "There is no good time to close this main link between Carluke and the Clyde Valley but doing so during the school summer holidays will hopefully minimise disruption as much as possible.
"Unfortunately vital resurfacing works on main routes cannot be completed without some disruption to road users.
"We apologise in advance for any inconvenience to the public and advise them to use the signed alternative routes for the duration of the works."
An alternative route has been put in place. For motorists travelling north along the A73 Kirkton Street, they should continue north on the A73 Stewart Street/Airdrie Road, then turn left onto B7011 Wildman Road, continue onto Station/Brownlee Road and finally left onto Mauldslie Road. Vehicles travelling in the opposite direction should follow the diversion vice versa.
