Road repairs are scheduled to take place on the A71 Stonehouse Road between Strathaven and Muirburn Road.
This is due to essential road surfacing work needing to be carried out.
South Lanarkshire Council has announced the road will be closed from 1am on 30 May until 12am on 31 May.
Councillor Chris Thompson said: "These works are essential to improve the road surface at this location."
The diversion routes will be clearly signposted.
From the Strathaven side, the alternative route will be via the A723 Strathaven to Hamilton, A72 to Cornsilloch Roundabout then A71 to Stonehouse.
From the Stonehouse side, the diversion will be vice-versa.
